I agree you should try the American directly into the amp - both into the front and also try it into the fx loop return.
The way you have it currently set-up is you have two solid state pre-amps into a digital pre-amp into a tube pre-amp into a tube power-amp. Lot of pre-amps at work there!

I have never ever played any of my amps without at least some Reverb on it because dry it just sounded bad but this American Sound into the FX Loop of this Bugera V55 made SUCH HUGE difference in amp tone Im awe struck! I have a "genuine" Fender tube amp folks! I played it for the last hour totally Dry and my oh my what nice harmonics come out of my notes especially chords! Just wow! Im buying you a pint next time Im in Ireland! What a great sounding amp I now have! Also the amp seems to be getting MUCH more Volume this way! Then I hit the front end of the amp with my NUX OD and man what nice creamy drive I get this way! I really need to think this one through and use the FX Loop to its full capacity placing all my Reverbs, Delays, Tremolo etc ... there! But this FX Loop is sensitive to noise so must take extra focus on getting a good Power Supply! HB has this new (came out this month) Pro ISO power supply. Im waiting to read the first reviews before I jump in to get it!

You would expect more volume and more noise in the FX loop Che. It's normal. The FX loop is often more accepting of line level signals so a big boost from the AS pedal is going to do that.
I use mine that way as my Fender amp has a solid state preamp into 6L6 power valves so I'm not getting lovely breakup tones from any preamp valves. The AS pedal does that for me instead.
Wonder why the V55 preamp valves aren't breaking up nicely. What are they?

You can see it on one of my photos in this thread; I think this amp has 2x6L6 and 3x12ax7
It does break up but with much less dynamics and sounds harsh. Driving the power section sure opens up some nice tone. Pity I don't have 4x10" to give it go with this setup!

Ok are you saying that the 12AX7A is in my Preamp Section??? I thought those 6L6 were in the Preamp section??? Good to know that I have best of the best So far Im very happy with Behringer U-Phoria UM2 audio interface and their BugeraV5 and V55 amps! Im glad TC Electronic was sold to Behrniger (and not some other company) because they seem to be aiming for affordable but quality gear for barefoot folks like myself!

Here is a short recording of the Bugera V55 with a American Sound into the FX loop. ST-62MN and TE-70BP
First recording is Ironstone Silver Strat pickups, second recording Roswell Tele pickups. Bugera V55 amp (all stock), American Sound pedal into FX Loop. NUX OD into the front of the amp. Before OD is a HB Compressor pedal. Dry signal. Touch of Stereo in Reaper DAW. No extra EQ! Cheap SM57 clone mic called Chord.

You have Guitar->Compressor->Nux OD->Bugera 12AX7 preamp->American Sound->Bugera 6L6 Poweramp You could also try Guitar->Compressor->Nux OD->American Sound->Bugera 6L6 Poweramp or even just Guitar->American Sound->Bugera 6l6 Poweramp

So to plug the guitar into the FX Loop basically you are saying Thats just plain strange but hey just might be the thing Im after. ... even to think about not plugging my guitar into the front of the amp is just make me feel dizzy feels like Im loosing the ground under my feet, as if I just landed on Mars Will have to try that!
