Wait! How do I plug the guitar into the FX Loop? I mean I have the American Sound in the Send AND Return! Where do you plug in the guitar???

Guitar into American Sound in, American Sound Out into the FX loop return. Try it but start with the volume of the American Sound low. Anyway Mick and Dan discuss the concept here - skip to 5.20. I haven't watched it all - those two lads go on a bit too long for my patience levels..

And from the Tech 21 Character pedal manual (The Joyo/HB is a clone of it)
APPLICATIONS AS A STOMPBOX with a guitar amp. You can connect the Character Series pedals in- line just as you would a standard distortion or overdrive pedal. If the pre-amp of your guitar amplifier is imparting too much of its own character on the pedal, plug into the low level input and set the pre-amp as clean and neutral as possible. Since most amps tend to be on the bright side, you may need to start with High below 12 o’clock and then adjust as necessary.You can also plug the pedal into the effects loop return (if the amp has one).This will disable the entire pre- amp of the amplifier allowing you to hear a truer representation of the sound. NOTE: the master volume on your amplifier will be defeated, so be sure to have the volume control down on the pedal.

Oh yeah lots more clarity and attack when going into the Return. Just played the guitar into American Sound into Return and it sounds awesome! Will try tomorrow placing the compressor, OD and Mooer Recho into the chain same way into Return. But I have noticed this way I cant use the amps EQ nor any of the knobs it seems! Well, AS has enough EQ for that as so does my NUX OD

On most amps EQ is part of the preamp.

Ok so preamp IS the Tone of the amp and power section is the 55 watts in this case. By going into Return I bypass the entire amps Tone section and use the True Tone pedal to get the Fender amp tone. How cool! Never thought I will like the FX loop so much! Must experiment more ...

That's it Che. That's what I do to avoid using the onboard preamp on the Fender 25SE, just use the American Sound as an outboard preamp instead. That's why I think of it as the eq controls of a Fender Deluxe in a box.
Don't know where your reverb is situated in your signal chain but it always seems to come straight after the preamp and before the send so you may lose your reverb control too. Seems a shame on my amp 'cos I love its reverb. If I know I'm going to be using it I take the AS pedal out of the fx loop altogether. It seems to sound better as a preamp BEFORE my own amp's preamp rather than after it. It all depends on what tone you want coloured by what.

Not even one knob can be used when in Return not even Reverb! But I don't use this reverb as its really bad!!! My Infinium reverb is actually awesome. Need to get me some TC Reverb !

Putting a reverb pedal (or delay with short delay) between the AS and the FX loop return is probably the way to go.

After messing about with all the different options suggested here I seem to prefer this way the most: guitar-NUX OD-Digitech RP225 into front of the amp then American Sound into FX loops Send-Return. Gives such a tight and rich tone.
So I have sim pedal hitting Bugera's preamp section and a sim pedal hitting the power section afterwards! Noise is not issue so this might well be the way to go and the RP225 has some fine sounding Drip Spring Reverb.
Now I need to test how the Mooer Recho delay sounds between the AS and FX Loop ...

Saw a video by TC Electroic were they play a strat into a Bugera V55 and tried their amp settings and voila HUGE improvement! The Bass knob is full on! This amp can be boomy but not if you crank the bass all the way up and then set the Mids to 6 and Treble to 6. They are like me in the Bright input and Clean channel. They keep the Master at 4 and Clean volume at 6. Yes! Sounds very good on its own but awesome with pedals (Lunar Drive). Can't wait to plug my new CKK Gears compressor into this new setting!
